BioGnost’s Hem Diff Strong is a strong differentiation reagent used in regressive hematoxylin-eosin (HE) staining (using Hematoxylin H), in both automated and manual staining procedures. Regressive staining refers to a method that includes overstaining structures and subsequent removal of excess dye from the cytoplasm, differentiation of nuclei with a strong differentiator, and better binding of eosin. The use of a strong differentiator in regressive staining is also necessary due to the removal of excess blue dye from the slides, a phenomenon that occurs mostly due to the vertical position of the slides during HE staining in automatic stainers where the specimens are immersed in a reagent container. Usually, a very strong differentiation reagent such as Acid alcohol enables rapid differentiation (within a few seconds), but such differentiation is not as precise as with the Hem Diff Strong reagent, which contains a lower concentration of acid that allows for a longer incubation (10 seconds). Regardless of the fixative in which the tissue is fixed (formalin or glyoxal), Hem Diff Strong gives clear results. The time spent for differentiation with the Hem Diff Strong reagent is the same regardless of the fixative used to fix the examined sample.
