Introduction
BioGnost’s BioBluing Buffer is a buffered alkaline reagent for nuclear bluing used in both automated and manual hematoxylin-eozin (HE) staining methods. Due to its low pH value, hematoxylin stains the nuclei red, and thanks to the process of bluing, which is carried out in an alkaline solution, the nuclei take on a characteristic blue color. The common use of tap water as a bluing reagent has many disadvantages, such as the non-uniform pH of the water, which varies very often. Other bluing reagents (based on carbonates and sulfates) do not contain a buffer and are therefore also subject to a large change in pH, which is most evident when staining a large number of slides, and consequently results in uneven staining. The BioBluing Buffer contains a buffer that keeps the pH constant (8.5) and thus enables uniform bluing of the nuclei and an optimal staining result. The solution is tinted blue for easier identification of the reagent.
